If you doing the Boss Quest (Poirot - 48th Floor Dimensional Gap Investigation or Poirot - 48th Floor Dimensional Gap Reinvestigation), wait until the cooldown of the boss floor crumbling, you'll automatically teleport to 49th floor and finish the quest by talking with Poirot there.
If you take the portal at the bottom of the stairway, you'll go back to Tower of Chaos 48th Floor. If you completed your Monster Quest, Poirot will not teleport you to either 48F Chaotic Space or Tower of Chaos 49th Floor (Safe Floor), so you'll have to cancel your Monster Quest to either redo it or teleport to Tower of Chaos 49th Floor (Safe Floor).
